No, whole blood is not applicable as input sample material on the Tapestri Platform. Cell types other than lymphocytes, such as erythrocytes, need to be removed
Single cells must be isolated from whole blood prior to usage on the platform. A number of commercially available isolation protocols exist, most of them gradient-based (Ficoll-Paque):
In general, the Tapestri Platform requires either single-cell or single-nuclei suspension with the following specifications:
Single-cell suspension
- Viable – 80 % as measured with Trypan blue
- Free of doublets, multiplets, and clumps
- Free of debris
- Not under/over-concentrated – ~3,000 cells/µL in a minimum of 35 µL volume
Single-nuclei suspension
- Intact and not partially digested nuclei as assessed by fluorescent microscopy
- Free of doublets, multiplets, and clumps
- Free of debris
- Not under/over-concentrated – ~3,000 nuclei/µL in a minimum of 35 µL volume
